This study investigates how hypoxia induces a "hypoxic memory" in breast cancer cells, leading to prolonged suppression of type I interferon (IFN) signaling and antigen presentation machinery. This suppression persists even after returning to normal oxygen levels, enhancing tumorigenic and metastatic abilities. The histone deacetylase inhibitor entinostat can reverse this hypoxic memory, suggesting potential therapeutic strategies to combat metastasis in breast cancer.

Marain NF et. al. (July 2024). Combined Exercise and Diet Induce Airway Hyperreactivity While Reducing Liver Steatosis in Mice with Diet-Induced Obesity Nutrients. 16(13)
This study explores the effects of combined exercise and diet on airway hyperreactivity and liver steatosis in mice with diet-induced obesity. After 27 weeks on a high-fat diet (HFD), mice were split into groups receiving either HFD or normal chow diet (NCD) for three weeks, with half of each group undergoing sub-maximal exercise. Results showed that the combined NCD and exercise regimen induced airway hyperreactivity and increased lung inflammation. However, this regimen also led to reduced body weight, hepatic steatosis, and fibrosis. These findings highlight the complex effects of combined exercise and diet on respiratory and liver health in obesity.

Lyu TJ et. al. (July 2024). DNMT3A dysfunction promotes neuroinflammation and exacerbates acute ischemic stroke MedComm (2020). 5(7):e652.
This study examines how DNMT3A dysfunction affects neuroinflammation and acute ischemic stroke (AIS) outcomes. Analyzing 8,524 ischemic stroke patients, researchers found that DNMT3A-driven clonal hematopoiesis of indeterminate potential (CHIP) is linked to greater neurological disability. Using a mouse model of stroke, they showed that DNMT3A protein levels rise in the brain's penumbra after stroke. Inhibiting DNMT3A with RG108 increased neutrophil proliferation, infiltration, and proinflammatory activation, worsening infarct volume and neurobehavioral outcomes. These findings indicate that DNMT3A mutations exacerbate stroke severity through heightened inflammation, suggesting new avenues for research and potential treatments.

Ganguly A et. al. (July 2024). Gestational exposure to air pollutants perturbs metabolic and placenta-fetal phenotype Reprod Toxicol. 128:108657.
This study investigates how gestational exposure to air pollution (AP) affects maternal and fetal health. Pregnant mice exposed to AP showed cardiovascular issues, hyperglycemia, glucose intolerance, and insulin resistance. Their placentas exhibited inflammation, cellular damage, and altered nutrient transporters, leading to reduced placental and fetal weights and decreased brain cortical thickness in fetuses. AP exposure also increased placental DNA 5'-hydroxymethylation and disrupted gene expression. These findings suggest that AP exposure during pregnancy mimics gestational diabetes mellitus and adversely affects placental and fetal development.

Ragot H et. al. (July 2024). Citrullinated Histone H3, a Marker for Neutrophil Extracellular Traps, Is Associated with Poor Prognosis in Cutaneous Squamous Cell Carcinoma Developing in Patients with Recessive Dystrophic Epidermolysis Bullosa Cancers (Basel). 16(13)
This study explores the role of citrullinated histone H3, a marker for neutrophil extracellular traps (NETs), in the prognosis of cutaneous squamous cell carcinoma (SCC) in patients with recessive dystrophic epidermolysis bullosa (RDEB). Analyzing 38 SCCs from 20 RDEB patients, researchers found that high-risk primary SCCs had higher neutrophil-to-lymphocyte ratios and increased NETs in the tumor microenvironment. Citrullinated histone H3 levels were elevated in the serum of patients with high-risk SCCs, suggesting it as a potential blood marker for poor prognosis in RDEB-SCCs. These findings highlight the significance of the tumor microenvironment and NETs in the progression of SCC in RDEB patients.

Wang K et. al. (July 2024). BUB1 potentiates gastric cancer proliferation and metastasis by activating TRAF6/NF-κB/FGF18 through m6A modification Life Sci. :122916.
This study investigates the role of BUB1 in gastric cancer (GC) proliferation and metastasis. Using in vitro and in vivo models, researchers found that BUB1 promotes GC cell growth and spread, with high BUB1 and FGF18 expression linked to poor patient prognosis. Mechanistically, BUB1 interacts with METTL3 to induce m6A modification of TRAF6 mRNA, activating the NF-κB/FGF18 pathway. BUB1 knockdown inhibited GC cell proliferation and metastasis, while BUB1 overexpression had the opposite effect. These findings highlight BUB1 as a key regulator of GC progression through the TRAF6/NF-κB/FGF18 axis, suggesting it as a potential target for GC diagnosis and therapy.

He S et. al. (July 2024). NtWIN1 regulates the biosynthesis of scopoletin and chlorogenic acid by targeting NtF6'H1 and NtCCoAMT genes in Nicotiana tabacum Plant Physiol Biochem. 214:108937.
This study explores the role of the ERF transcription factor NtWIN1 in regulating the biosynthesis of scopoletin and chlorogenic acid (CGA) in Nicotiana tabacum. NtWIN1, highly expressed in tobacco stems, sepals, and pistils, was found to influence the accumulation of these polyphenols. Overexpression of NtWIN1 increased CGA levels by 25%-50% but decreased scopoletin by 30%-67%, while NtWIN1 knock-out lines showed reduced CGA and increased scopoletin. NtWIN1 modulates the expression of NtF6'H1 and NtCCoAMT by binding to their promoters. The study identifies NtWIN1 as a key regulator of polyphenol synthesis in tobacco, providing potential targets for metabolic engineering in plant polyphenol production.

Xiao C et. al. (July 2024).  Overcoming Cancer Persister Cells by Stabilizing the ATF4 Promoter G-quadruplex Adv Sci (Weinh). :e2401748.
This study explores how stabilizing the G-quadruplex (G4) structure in the ATF4 promoter can overcome cancer persister cells, which are resistant to treatment. Glutamine restriction induces these persister cells, which activate the GCN2-ATF4-ASCT2 axis, leading to resistance. Researchers discovered that coptisine (COP) stabilizes the ATF4-G4 structure, disrupting its interaction with the transcription factor TFAP2A. This stabilization reduces ATF4 levels and induces cancer cell death. The findings suggest targeting the ATF4-G4 structure as a promising strategy for tackling treatment-resistant cancer cells.

This study investigates how remifentanil (RE) protects against hepatic ischemia/reperfusion injury (HIRI) by regulating the BACH1/PRDX1 axis. Using mouse and hepatocyte models, researchers found that RE improves liver function and reduces oxidative stress and apoptosis. RE treatment increased PRDX1 levels and decreased BACH1, which normally inhibits PRDX1. BACH1 binds to the PRDX1 promoter, suppressing its expression and contributing to oxidative stress. Overexpressing BACH1 or silencing PRDX1 counteracted RE's protective effects. These findings suggest that RE mitigates HIRI by modulating the BACH1/PRDX1 pathway, offering insights into its therapeutic potential.
