Western blot analysis of lysates from THP-1 cells using Cleaved Gasdermin D (N Terminal) mAb at 1:72000 dilution incubated overnight at 4°C. THP-1 cells were treated with PMA/TPA (80 nM) at 37°C for overnight and LPS (1 µg/ml) at 37°C for 6 hours.
Secondary antibody: HRP-conjugated Goat anti-Rabbit IgG (H+L) at 1:10000 dilution.
Lysates/proteins: 30 µg per lane.
Blocking buffer: 3 % nonfat dry milk in TBST.

Western blot analysis of lysates from wild type (WT) and Cleaved Gasdermin D knockout (KO) THP-1 cells using Cleaved Gasdermin D (N Terminal) mAb at 1:72000 dilution incubated at room temperature for 1.5 hours. THP-1 cells and Gasdermin D knockout (KO) THP-1 cells were treated with PMA/TPA (80 nM) at 37°C for overnight and LPS (1 µg/ml) at 37°C for 6 hours.
Secondary antibody: HRP-conjugated Goat anti-Rabbit IgG (H+L) at 1:10000 dilution.
Lysates/proteins: 30 µg per lane.
Blocking buffer: 3% nonfat dry milk in TBST.

Confocal imaging of THP-1 cells (treated with PMA and LPS) and THP-1 cells (untreated) cells using Cleaved Gasdermin D (N Terminal) mAb (dilution 1:200) followed by a further incubation with Cy3 Goat Anti-Rabbit IgG (H+L) (dilution 1:500) (Red). DAPI was used for nuclear staining (Blue). Objective: 100x.
