This study explores the effects of combined exercise and diet on airway hyperreactivity and liver steatosis in mice with diet-induced obesity. After 27 weeks on a high-fat diet (HFD), mice were split into groups receiving either HFD or normal chow diet (NCD) for three weeks, with half of each group undergoing sub-maximal exercise. Results showed that the combined NCD and exercise regimen induced airway hyperreactivity and increased lung inflammation. However, this regimen also led to reduced body weight, hepatic steatosis, and fibrosis. These findings highlight the complex effects of combined exercise and diet on respiratory and liver health in obesity.

This study examines how DNMT3A dysfunction affects neuroinflammation and acute ischemic stroke (AIS) outcomes. Analyzing 8,524 ischemic stroke patients, researchers found that DNMT3A-driven clonal hematopoiesis of indeterminate potential (CHIP) is linked to greater neurological disability. Using a mouse model of stroke, they showed that DNMT3A protein levels rise in the brain's penumbra after stroke. Inhibiting DNMT3A with RG108 increased neutrophil proliferation, infiltration, and proinflammatory activation, worsening infarct volume and neurobehavioral outcomes. These findings indicate that DNMT3A mutations exacerbate stroke severity through heightened inflammation, suggesting new avenues for research and potential treatments.
