BackgroundSMARCAL1 is a subunit of the SWI/SNF chromatin remodeling complex. The SWI/SNF family members are known to display helicase and ATPase activities and regulate transcription of certain genes by altering chromatin structure. The protein encoded by this gene has sequence similarity to the E. coli RNA polymerase-binding protein HepA. Defects in this gene increase the risk of Schimke immuno-osseous dysplasia (SIOD), which is a rare autosomal recessive disorder with the diagnostic features of spondyloepiphyseal dysplasia, renal dysfunction, and T-cell immunodeficiency. SMARCAL1 mutations affect protein activity, protein stability, or the protein's ability to bind to chromatin.
