BackgroundSpecific phosphatidylinositol (PtdIns) phosphatase targeting PtdIns(3,4,5)P3, converting it to PtdIns(3,4)P2, which dampens PI3K pathways. Negatively regulates B-cell antigen receptor and FC-gamma-RIIB signaling, terminating immune receptor transduction. Governs myeloid cell processes, mast cell degranulation, immune cell homeostasis, platelet signaling, and B-cell JNK signaling. Influences osteoclast precursor proliferation, macrophage behavior, phagocytosis, and endotoxin tolerance. Controls cell-cell junctions, neutrophil migration, CD32a signaling, and EGF-induced phospholipase C activity. Vital for neutrophil chemotaxis, FCGR3/CD16-mediated NK cell cytotoxicity, and activin/TGF-beta-induced apoptosis via Smad-dependent expression. Could also affect higher inositol polyphosphate levels like InsP6 through PtdIns(1,3,4,5)P4 hydrolysis.
