BackgroundEssential serine/threonine-protein kinase in mitosis with two vital roles: signaling spindle-assembly checkpoint and ensuring accurate chromosome alignment. Facilitates checkpoint proteins' kinetochore assembly, influencing various factors like CENPF, BUB1B, and PLK1. Also involved in centromeric enrichment of AUKRB and SGO1 for sister chromatid cohesion. Interacts with APC/C-Cdh1 to regulate chromosome segregation and phosphorylates BUB3. Inhibits APC/C and CDC20 during spindle-assembly checkpoint activation. Facilitates accurate chromosome alignment but not the main role in spindle-assembly checkpoint. Induces cell death upon chromosome missegregation and suppresses spontaneous tumorigenesis.
