BackgroundKey component of MAP kinase pathway. Ligand binding (growth factors, cytokines) activates RAS and initiates RAF1 activation. RAF1 further activates MAP2K1/MEK1 and MAP2K2/MEK2. Both MEK1 and MEK2 phosphorylate MAPK3/ERK1 and MAPK1/ERK2, leading to signal transduction. This cascade regulates cell growth, survival, differentiation, transcription, metabolism, and cytoskeletal changes. Targets include PPARG, influencing differentiation and apoptosis. Also affects endosomal dynamics and Golgi fragmentation during mitosis.
