It collaborates with co-chaperones, influencing substrate recognition, ATPase cycle, and chaperone function. Co-chaperones also connect it to various client protein types, acting as adapters. The cycle starts with ATP and co-chaperone recruitment, followed by client protein binding, forming a functional chaperone. Once the client protein is properly folded, it and the co-chaperone leave the HSP90 chaperone in a partially open, ADP-bound state, prepared for the next cycle. Beyond chaperone activity, it affects transcription machinery regulation at multiple levels. It impacts transcription factor levels, modifies epigenetic activity, and participates in histone eviction, activating gene expression.
