This study investigates how epigenetic mechanisms regulate blood pressure (MAP) and kidney function in male and female Npr1 haplotype mice. Using the HDAC inhibitor mocetinostat (MGCD), the researchers treated mice with different copies of the Npr1 gene. Male mice with one copy (Npr1+/-) had higher MAP, kidney dysfunction, and fibrosis compared to those with two or three copies. MGCD treatment improved blood pressure and kidney function, showing different effects in males and females. The study found that epigenetic changes, such as histone modifications and microRNA expression, play a key role in hypertension and kidney issues, with notable sex-specific differences.

Xue X et. al. (August 2024). 17β-estradiol promotes the progression of temporomandibular joint osteoarthritis by regulating the FTO/IGF2BP1/m6A-NLRC5 axis Immun Inflamm Dis. 12(8):e1361.
This study examines how 17β-estradiol (E2) promotes temporomandibular joint osteoarthritis (TMJOA) progression through the FTO/IGF2BP1/m6A-NLRC5 axis. In TNF-α-treated rat fibroblast-like synoviocytes (FLSs), E2 increased cell proliferation, migration, invasion, and inflammation. E2 downregulated FTO, raising m6A methylation and stabilizing NLRC5 via IGF2BP1. In a TMJOA rat model, E2 worsened joint pathology and bone density, but NLRC5 knockdown alleviated these effects. Thus, E2 promotes TMJOA by modulating this molecular pathway.

Ding PF et. al. (July 2024). miR-93-5p impairs autophagy-lysosomal pathway via TET3 after subarachnoid hemorrhage Exp Neurol. 380:114904.
This study explores how miR-93-5p impairs the autophagy-lysosomal pathway (ALP) via TET3 after subarachnoid hemorrhage (SAH). Following SAH, the ALP is disrupted, with reduced autophagic flux and increased autophagosomes, alongside decreased TET3 expression. Activating TET3 improves ALP and neural function, while silencing TET3 worsens ALP function and increases apoptosis. Elevated miR-93-5p after SAH suppresses TET3, suggesting miR-93-5p as a potential target for neuroprotective therapy. This study highlights TET3's crucial role in ALP impairment post-SAH.

Kanojia A et. al. (August 2024). Interplay between DOT1L and HDAC1 regulates Leishmania donovani infection in human THP-1 cells Acta Trop. :107352.
This study investigates how DOT1L and HDAC1 regulate Leishmania donovani infection in human THP-1 cells. Leishmania donovani, which causes visceral leishmaniasis, upregulates HDAC1, leading to repression of host defensin genes. The researchers found that DOT1L, a histone methyltransferase, is also upregulated during infection, increasing H3K79 methylation. DOT1L regulates HDAC1 expression, and reducing DOT1L decreases HDAC1, increases defensin gene transcription, and lowers parasite load. Conversely, HDAC1 regulates DOT1L expression. This feedback loop between DOT1L and HDAC1 downregulates defensin genes during infection.

Yang L et. al. (August 2024).  Anticancer effects of Erzhimaoling decoction in high-grade serous ovarian cancer in vitro and in vivo Yang L et. al. (August 2024). 
This study explores the anticancer effects of Erzhimaoling decoction (EZMLD) on high-grade serous ovarian cancer (HGSOC) both in vitro and in vivo. EZMLD-containing serum, derived from rats, reduced SKOV3 ovarian cancer cell viability and increased apoptosis. The treatment upregulated METTL3/14 and FAS-mediated apoptosis proteins, while downregulating Keratin 8 (KRT8) and increasing KRT8 mRNA m6A methylation. Overexpression of METTL3/14 enhanced EZMLD’s effects, while knockdown reduced them. In vivo, EZMLD inhibited SKOV3 xenograft growth and induced apoptosis, indicating its potential as a therapeutic option for ovarian cancer and possibly other cancers.

Wu L et. al. (August 2024). Luoshi Neiyi Prescription inhibits estradiol synthesis and inflammation in endometriosis through the HIF1A/EZH2/SF-1 pathway J Ethnopharmacol. :118659.
This study investigates how Luoshi Neiyi Prescription (LSNYP) affects endometriosis (EMS) by targeting the HIF1A/EZH2/SF-1 pathway. LSNYP, containing 185 identified components, reduced estradiol (E2) and inflammation in endometrial cells. Hypoxia increased E2 and decreased EZH2, while LSNYP inhibited E2 production, cell proliferation, and inflammation. In vivo, LSNYP decreased inflammation and disrupted the HIF1A/EZH2/SF-1 pathway in endometriotic tissues, highlighting its potential as an EMS treatment.

Guo M et. al. (August 2024). Loss of cold tolerance is conferred by absence of the WRKY34 promoter fragment during tomato evolution Nat Commun. 15(1):6667.
This study investigates how tomato evolution has affected cold tolerance. By analyzing ATAC-Seq and RNA-Seq data, researchers found that a 60 bp InDel in the WRKY34 promoter is linked to differences in cold tolerance between wild and cultivated tomatoes. This fragment includes a GATA cis-regulatory element that suppresses WRKY34 expression under cold stress. WRKY34 also impacts the CBF cold response pathway. The findings highlight the role of promoter polymorphisms in gene expression and crop evolution.
