This study investigates how TNF-α influences pulmonary artery smooth muscle cell (PASMC) proliferation and the underlying mechanisms. TNF-α treatment increased superoxide production, NADPH oxidase activity, and PASMC numbers. It also caused DNA hypomethylation and reduced H3-K4 histone methylation. DNA methylation changes were linked to increased GADD45-α expression. Superoxide scavenger tempol mitigated these effects. TNF-α did not affect histone acetylation but implicated LSD1 in histone demethylation. The findings suggest TNF-α-induced PASMC proliferation involves excessive superoxide formation and epigenetic alterations.

Tang YW et. al. (May 2024). Triptolide decreases podocytes permeability by regulating TET2-mediated hydroxymethylation of ZO-1 Exp Biol Med (Maywood). 249:10051.
This article examines how triptolide (TP) affects podocyte permeability via TET2-mediated regulation of ZO-1 hydroxymethylation. TP upregulates ZO-1 expression and reduces podocyte permeability in a puromycin aminonucleoside (PAN)-induced injury model. TP also increases the DNA demethylase TET2, which enhances ZO-1 expression by decreasing promoter methylation. Inhibitors of DNA methylation mimic TP's effects, while TET2 knockdown increases methylation and podocyte permeability. The findings suggest TP alleviates podocyte permeability through TET2-mediated hydroxymethylation of ZO-1.

Shorey-Kendrick LE et. al. (June 2024). Development and Validation of a Novel Placental DNA Methylation Biomarker of Maternal Smoking during Pregnancy in the ECHO Program Environ Health Perspect. 132(6):67005.
This study develops placental DNA methylation biomarkers for maternal smoking during pregnancy (MSDP). Using machine learning, logistic LASSO regression showed the best performance in identifying DNAm signatures. The models were validated with high accuracy in external datasets. A key CpG site (cg27402634) was accurately measured by digital PCR. These biomarkers can effectively classify MSDP and offer a cost-effective screening method.

Ren M et. al. (June 2024). KIAA1429 promotes gastric cancer progression by destabilizing RASD1 mRNA in an m6A-YTHDF2-dependent manner J Transl Med. 22(1):584.
This article explores KIAA1429's role in gastric cancer (GC). Elevated in GC tissues, KIAA1429 is linked to poor prognosis. KIAA1429 promotes GC cell proliferation, migration, and invasion in vitro and enhances tumor growth in vivo. Mechanistically, KIAA1429 increases m6A methylation of RASD1 mRNA, leading to its destabilization via an m6A-YTHDF2-dependent pathway, thereby reducing RASD1 expression. RASD1 knockdown partially rescues the effects of KIAA1429 knockdown. KIAA1429 may serve as a prognostic biomarker and therapeutic target in GC.

Wysong BC et. al. (June 2024). HUMAN CST STIMULATES BASE EXCISION REPAIR TO PREVENT THE ACCUMULATION OF OXIDATIVE DNA DAMAGE J Mol Biol. :168672.
This study explores the role of the CTC1-STN1-TEN1 (CST) complex in DNA repair. CST, known for telomere maintenance, also stimulates base-excision repair (BER) proteins like OGG1, Pol β, APE1, and LIGI, enhancing their activity on telomeric and non-telomeric DNA. Biochemical assays confirm CST's role in promoting BER. CST knockout leads to increased 8-oxoguanine levels, indicating defective BER. This identifies CST as a key factor in preventing oxidative DNA damage by enhancing BER efficiency, highlighting its broader role in maintaining genome integrity.

Wu S et. al. (June 2024). H3K18 lactylation accelerates liver fibrosis progression through facilitating SOX9 transcription Exp Cell Res. 440(2):114135.
This study reveals that H3K18 lactylation promotes liver fibrosis by enhancing SOX9 transcription. Using liver fibrosis models, researchers found increased H3K18 lactylation in fibrotic livers and activated hepatic stellate cells (HSCs). Reducing lactylation through LDHA knockdown improved fibrosis by inhibiting HSC activity. Chromatin immunoprecipitation and luciferase assays confirmed that H3K18 lactylation activates SOX9, driving fibrosis progression. This discovery suggests targeting H3K18 lactylation as a potential therapeutic strategy for liver fibrosis.

Hou X et. al. (June 2024). KCNK1 promotes proliferation and metastasis of breast cancer cells by activating lactate dehydrogenase A (LDHA) and up-regulating H3K18 lactylation PLoS Biol. 22(6):e3002666.
This article identifies KCNK1 as a key promoter of breast cancer proliferation and metastasis. KCNK1 is upregulated in breast cancer and linked to poor prognosis. It enhances cancer cell growth and spread by activating lactate dehydrogenase A (LDHA), increasing glycolysis and lactate production. This activation leads to histone lactylation, which upregulates genes that further boost LDHA expression. This positive feedback loop reduces tumor cell stiffness and adhesion, driving cancer progression. KCNK1 is proposed as a potential biomarker and therapeutic target for breast cancer treatment.
