BackgroundPro-apoptotic kinase activated by stress and caspase-cleavage, leading to nuclear entry and chromatin changes, including DNA fragmentation. Crucial part of the Hippo pathway, controlling organ size and tumor suppression by limiting proliferation and promoting apoptosis. The pathway involves a kinase cascade: STK3/MST2 and STK4/MST1 complexed with SAV1 activate LATS1/2 complexed with MOB1, ultimately inhibiting YAP1 and WWTR1/TAZ oncogenes. This inhibits their roles in gene regulation linked to proliferation, cell death, and migration. STK3/MST2 and STK4/MST1 also prevent hepatocyte proliferation, stem cell activation, and tumor growth. Phosphorylates NKX2-1, NEK2, and others for centrosome function, transcriptional activation, and RAF1 activity. Co-activates ESR1 with SAV1 and enhances STK38 with MOBKL1B.
