BackgroundTyrosine-protein kinase functioning as a cell-surface receptor for fibroblast growth factors, critically regulating cell proliferation, differentiation, migration, apoptosis, and embryonic development. Essential for embryonic patterning, trophoblast function, limb and lung development, osteogenesis, and skin formation. A key player in osteoblast regulation, influencing differentiation, proliferation, and apoptosis. Also influences keratinocyte proliferation and osteoblast apoptosis. Phosphorylates PLCG1, FRS2, and PAK4, activating diverse signaling pathways. Activation of PLCG1 produces diacylglycerol and inositol 1,4,5-trisphosphate. FRS2 phosphorylation leads to cascades involving RAS, MAP kinases, AKT1, and more. FGFR2 signaling is controlled via ubiquitination, internalization, and degradation. Mutations disrupting FGFR2 maturation or activation result in aberrant signaling. Overexpressed FGFR2 promotes STAT1 activation.
