Activity AssayActivity of recombinant TET1 protein was measured with the Epigenase 5mC-Hydroxylase TET Activity/Inhibition Assay Kit (Fluorometric). In this assay, a TET substrate is converted to hydroxymethylated products, which is recognized with a specific antibody. The ratio or amount of hydroxymethylated products is proportional to enzyme activity and can then be fluorometrically measured at 530ex/590em. The activity of the TET enzyme is in turn proportional to the relative fluorescent units measured.
