BackgroundThis key regulator controls cell division entry and functions as a tumor suppressor. When phosphorylated by CDK3/cyclin-C, it promotes G0-G1 transition. It represses E2F1 target genes' transcription, maintaining cell cycle arrest by interacting with E2F1 in its underphosphorylated form. It aids heterochromatin formation by stabilizing histone methylation, particularly in constitutive heterochromatin, through histone methyltransferase recruitment. It inhibits TAF1's kinase activity, represses c-FOS transcription with SMARCA4/BRG1, and mediates transcriptional repression upon viral infections. Viral proteins like SV40 large T antigen, HPV E7, or adenovirus E1A disrupt RB1-E2F1 complex, altering RB1's activity.
