This study examines how endothelial cell-derived extracellular vesicles (EC-EVs) affect mesenchymal stem cell (MSC) therapy for acute respiratory distress syndrome (ARDS). Using a lung injury model, researchers discovered that EC-EVs from ARDS patients reduced MSC viability and migration by decreasing isocitrate dehydrogenase 2 (IDH2) levels. This reduction disrupted the IDH2/TET pathway, leading to a decrease in α-ketoglutarate (α-KG) and worsening lung injury. The findings suggest that EC-EVs in the lung microenvironment hinder MSC therapy through the IDH2/TET pathway, providing insights for improving MSC-based ARDS treatments.

This is mediated by increased interactions between Sp1, β-catenin, and p300 on target gene promoters. NR2E3 maintains epigenetic homeostasis, suppressing Wnt/β-catenin signaling and HCC development.

ALKBH5 in the mPFC influenced depression behaviors bidirectionally, affecting the m6A modification of the glutamate transporter-1 (GLT-1), boosting its expression in astrocytes. Removing astrocytic ALKBH5 protected against stress-induced disruptions in glutamatergic synaptic transmission, neuronal atrophy, and defective Ca2+ activity. Enhancing m6A modification with S-adenosylmethionine (SAMe) also had antidepressant-like effects. Targeting astrocytic ALKBH5 could be a potential therapy for depression.
