Wu X et. al. (May 2024). Endothelial cell-derived extracellular vesicles modulate the therapeutic efficacy of mesenchymal stem cells through IDH2/TET pathway in ARDS Cell Commun Signal. 22(1):293.
This study examines how endothelial cell-derived extracellular vesicles (EC-EVs) affect mesenchymal stem cell (MSC) therapy for acute respiratory distress syndrome (ARDS). Using a lung injury model, researchers discovered that EC-EVs from ARDS patients reduced MSC viability and migration by decreasing isocitrate dehydrogenase 2 (IDH2) levels. This reduction disrupted the IDH2/TET pathway, leading to a decrease in α-ketoglutarate (α-KG) and worsening lung injury. The findings suggest that EC-EVs in the lung microenvironment hinder MSC therapy through the IDH2/TET pathway, providing insights for improving MSC-based ARDS treatments.

Ingelson-Filpula WA et. al. (May 2024). Hypoxia-induced downregulation of RNA m6A protein machinery in the naked mole-rat heart Biochimie. 225:125-132.
The article explores how hypoxia affects RNA m6A protein machinery in the hypoxia-tolerant naked mole-rat heart. Researchers exposed Heterocephalus glaber to low oxygen for 24 hours and measured m6A methylation-related proteins, demethylase activity, and total m6A content in cardiac tissue. While demethylase activity and total m6A levels remained unchanged, many m6A pathway proteins were downregulated during hypoxia. This suggests that m6A may not be a primary hypoxia response in the naked mole-rat heart, but the downregulation of m6A machinery indicates other biological roles. Further research will explore additional RNA modifications and epigenetic mechanisms to understand hypoxia tolerance in this species.

Zhao Y et. al. (May 2024).  Inhibition of endothelial histone deacetylase 2 shifts endothelial-mesenchymal transitions in cerebral arteriovenous malformation models J Clin Invest.
This study examines the role of endothelial histone deacetylase 2 (HDAC2) in cerebral arteriovenous malformations (AVMs), a leading cause of hemorrhagic strokes. Using mouse models, researchers discovered that cerebral endothelial cells (ECs) acquired mesenchymal markers, contributing to vascular malformations. Inhibiting HDAC2 prevented ECs from undergoing mesenchymal differentiation and reduced AVMs. They found altered expression of HDAC2 and enhancer of zeste homolog 1 (EZH1) in AVMs, affecting histone modifications (H4K8ac and H3K27me) and regulating EC differentiation. This study highlights the epigenetic mechanisms underlying AVMs, offering potential therapeutic targets.

Leung YK et. al. (May 2024). The Loss of an Orphan Nuclear Receptor NR2E3 Augments Wnt/β-catenin Signaling via Epigenetic Dysregulation that Enhances Sp1-β catenin-p300 Interactions in Hepatocellular Carcinoma Adv Sci (Weinh). :e2308539.
This study investigates the tumor-suppressive role of NR2E3 in hepatocellular carcinoma (HCC). Low NR2E3 expression in HCC patients is linked to poor outcomes and increased Wnt/β-catenin signaling. NR2E3 knockout mice show faster tumor growth and higher Wnt/β-catenin activity. NR2E3 loss enhances tumor aggressiveness and upregulates key Wnt/β-catenin pathway genes. This is mediated by increased interactions between Sp1, β-catenin, and p300 on target gene promoters. NR2E3 maintains epigenetic homeostasis, suppressing Wnt/β-catenin signaling and HCC development.

Zemanová V et. al. (May 2024). Multicontamination Toxicity Evaluation in the Model Plant Lactuca sativa L Plants (Basel). 13(10)
This study explores how arsenic, cadmium, lead, and zinc contamination affects lettuce. After 45 days, these contaminants significantly reduced root and leaf biomass, caused oxidative stress, and led to visible toxicity symptoms like leaf chlorosis and root browning. They also disrupted nitrogen and carbon metabolism, induced senescence, and decreased chlorophyll and carotenoid levels. Overall, the study confirms that combined contamination damages lettuce's antioxidant system and increases leaf sensitivity to toxins.

Guo F et. al. (May 2024). Astrocytic ALKBH5 in stress response contributes to depressive-like behaviors in mice Nat Commun. 15(1):4347.
This study links increased ALKBH5 in astrocytes to depressive behaviors. ALKBH5 levels rose in MDD patients and models, particularly in astrocytes under stress. Deleting ALKBH5 in astrocytes, but not in other cells, led to antidepressant-like effects. ALKBH5 in the mPFC influenced depression behaviors bidirectionally, affecting the m6A modification of the glutamate transporter-1 (GLT-1), boosting its expression in astrocytes. Removing astrocytic ALKBH5 protected against stress-induced disruptions in glutamatergic synaptic transmission, neuronal atrophy, and defective Ca2+ activity. Enhancing m6A modification with S-adenosylmethionine (SAMe) also had antidepressant-like effects. Targeting astrocytic ALKBH5 could be a potential therapy for depression.
