Wang R et. al. (June 2024). TET1 Regulates Nestin Expression and Human Airway Smooth Muscle Proliferation Am J Respir Cell Mol Biol.
This study investigates the role of TET1 in regulating nestin expression and human airway smooth muscle (ASM) proliferation, which are implicated in asthma. The researchers found that TET1 activity and protein were increased in asthmatic ASM cell cultures, along with higher levels of 5-hydroxymethylcytosine (5-hmC), a product of TET1 activity. Knockdown of TET1 reduced 5-hmC levels and nestin expression, a protein involved in cell proliferation through the mTOR pathway. Inhibition of TET1 also decreased PDGF-induced signaling and ASM cell proliferation. These findings suggest that TET1 contributes to ASM proliferation in asthma, potentially through the nestin-mTOR axis.

Gong Y et. al. (June 2024). Lysine acetyltransferase 14 mediates TGF-β-induced fibrosis in ovarian endometrioma via co-operation with serum response factor J Transl Med. 22(1):561.
This study explores how lysine acetyltransferase 14 (KAT14) contributes to fibrosis in ovarian endometrioma, driven by transforming growth factor-β (TGF-β). KAT14 is upregulated in endometrioma lesions and promotes a fibrotic response in cells. In mice with endometriosis, reducing KAT14 levels lessens fibrosis. KAT14 works with serum response factor (SRF) to increase α-smooth muscle actin (α-SMA) expression and extracellular matrix production. Targeting KAT14 might offer a new way to treat endometrioma-related fibrosis.

Kawarai Y et. al. (June 2024). Alterations in DNA methylation machinery in a rat model of osteoarthritis of the hip J Orthop Surg Res. 19(1):357.
This study explores changes in DNA methylation-related genes and global methylation in a rat model of hip osteoarthritis (OA). Rats with induced OA showed OA symptoms and lower pain thresholds. In the OA group, genes related to DNA methylation (Dnmt 3a, Tet 1, Tet 3) were upregulated in synovial tissue, and global DNA methylation was higher compared to controls. These findings suggest altered DNA methylation machinery in hip OA.

Fu XL et. al. (June 2024). HBXIP induces PARP1 via WTAP-mediated m6A modification and CEBPA-activated transcription in cisplatin resistance to hepatoma Acta Pharmacol Sin.
This study explores how hepatitis B X-interacting protein (HBXIP) contributes to cisplatin resistance in hepatoma. HBXIP increases the expression of PARP1, a protein involved in DNA repair and cisplatin resistance. HBXIP does this by promoting a modification (m6A) in the PARP1 gene and activating a transcription factor (CEBPA) that enhances PARP1 expression. The findings suggest a potential role for aspirin in overcoming cisplatin resistance by inhibiting HBXIP and reducing PARP1 expression.

Sun Y et. al. (June 2024). The CsDof1.8-CsLIPOXYGENASE09 module regulates C9 aroma production in cucumber Plant Physiol.
This study investigates the regulation of C9 aroma production in cucumber fruits. Researchers identified Lipoxygenase09 (CsLOX09) as a key gene controlling C9 aroma accumulation. Higher CsLOX09 expression in one cucumber line led to more C9 aromas. Knocking out CsLOX09 reduced C9 aroma content significantly. They also discovered that a transcription factor, DNA binding with One Finger 1.8 (CsDof1.8), directly regulates CsLOX09 expression. Overexpressing CsDof1.8 increased C9 aroma content and CsLOX09 expression. These findings shed light on the metabolic regulation of C9 aromas in cucumber, aiding future efforts in cucumber breeding for improved flavor.

Feng H et. al. (June 2024). FTO alleviated ferroptosis in septic cardiomyopathy via mediating the m6A modification of BACH1 Biochim Biophys Acta Mol Basis Dis. :167307.
This study explores how FTO, a regulator of m6A methylation, affects septic cardiomyopathy, a condition involving heart dysfunction during sepsis. Researchers found that FTO overexpression in cell and animal models reduced ferroptosis (a type of cell death), lowered heart inflammation and dysfunction, and improved survival rates. However, these protective effects were reversed by overexpressing BACH1, a molecule negatively regulated by FTO. FTO controls the m6A modification of BACH1, suggesting a potential therapeutic target for sepsis-induced heart injuries.

Zhou L et. al. (June 2024). YTHDC1 inhibits autophagy-dependent NF-κB signaling by stabilizing Beclin1 mRNA in macrophages J Inflamm (Lond). 21(1):22.
This study investigates the role of YTHDC1, an m(6)A nuclear reader, in regulating inflammatory bowel disease (IBD). Using mouse models of colitis and in vitro macrophage inflammation models, researchers found that YTHDC1 levels were decreased in IBD. Overexpressing YTHDC1 reduced inflammatory gene expression and NF-κB activation in macrophages, while inhibiting YTHDC1 had the opposite effect. YTHDC1 was found to stabilize Beclin1 mRNA, promoting autophagy and inhibiting NF-κB signaling. These findings suggest that targeting YTHDC1 could be a potential therapeutic strategy for IBD.

Wu Z et. al. (June 2024).  TGF-β1 facilitates gallbladder carcinoma metastasis by regulating FOXA1 translation efficiency through m6A modification Cell Death Dis. 15(6):422.
This study explores how TGF-β1 contributes to gallbladder carcinoma (GBC) metastasis by affecting the translation of FOXA1 mRNA through m6A modification. Researchers found that TGF-β1 reduces FOXA1 expression, promoting GBC cell migration and EMT. They discovered that TGF-β1 increases m6A modification on the coding sequence (CDS) region of FOXA1 mRNA, inhibiting its translation. Additionally, they identified ALKBH5 as a regulator that counteracts this effect, promoting FOXA1 protein expression. These findings suggest a mechanism by which TGF-β1 promotes GBC metastasis through m6A modification of FOXA1 mRNA.
