This study investigates how nitric oxide (NO) regulates DNA methylation by inhibiting ten-eleven translocation (TET) DNA demethylases and ALKBH2. NO forms a dinitrosyliron complex (DNIC) with the enzymes’ iron cofactors, blocking their activity and leading to increased levels of 5-methylcytosine (5mC) and 5-hydroxymethylcytosine (5hmC) in cancer cells. This reveals a novel epigenetic role for NO, distinct from its canonical signaling pathways, with implications for gene regulation in tumor development.

Zhang BB et. al. (February 2025). Anti-fibrosis effect and its mechanism of atractylenolide III on post-traumatic extending knee joint contracture in rats Exp Gerontol. 201:112708.
This study explores the anti-fibrotic effects of atractylenolide III (ATL III) on post-traumatic knee joint contracture (PTJC) in rats. ATL III reduced fibrosis, inflammation, and collagen accumulation in rat knees while inhibiting synovial fibroblast proliferation and migration in vitro. Mechanistically, ATL III appears to target Sirt1, promoting Smad3 deacetylation and thereby alleviating knee fibrosis.

Shen T et. al. (February 2025). HIF2α drives ccRCC metastasis through transcriptional activation of methylation-controlled J protein and enhanced prolegumain secretion Cell Death Dis. 16(1):93.
This study investigates the role of HIF2α in promoting metastasis in clear cell renal cell carcinoma (ccRCC) through the transcriptional activation of methylation-controlled J protein (MCJ). HIF2α enhances prolegumain secretion by upregulating MCJ, which disrupts mitochondrial function and increases oxidative stress, contributing to tumor progression. Targeting MCJ or using ROS scavengers like MitoQ effectively reduced prolegumain secretion, mitochondrial damage, and lung metastasis in a ccRCC mouse model.

This study examines how the core circadian clock component CCA1 regulates the unfolded protein response (UPR) during endoplasmic reticulum (ER) stress in plants. CCA1 enhances the expression of UPR target genes by forming a complex with the UPR modulator bZIP28, promoting ER stress recovery. This time-of-day-dependent regulation highlights the role of the circadian clock in optimizing ER proteostasis and stress tolerance.

Guan H et. al. (February 2025). Upregulation of YY1 in M2 macrophages promotes secretion of exosomes containing hsa-circ-0000326 via super-enhancers to facilitate prostate cancer progression Mol Cell Biochem.
This study explores how upregulation of the transcription factor YY1 in M2 macrophages promotes prostate cancer (PCa) progression. YY1 enhances the secretion of exosomes containing hsa-circ-0000326 by activating super-enhancers of the MALAT1 gene and upregulating QKI expression, which drives circRNA formation. These exosomes facilitate PCa metastasis by increasing androgen receptor levels, highlighting hsa-circ-0000326 as a potential therapeutic target.

Abulehia H et. al. (February 2025). Intake of Trans-Fat Diet With Prenatal Bisphenol A Exposure Induce Global DNA Methylation in Rats Offspring Birth Defects Res. 117(2):e2450.
This study investigates the combined effects of prenatal Bisphenol A (BPA) exposure and postnatal trans-fat diet (TFD) intake on global DNA methylation in rat offspring. While no changes were observed in PPAR-γ protein expression, the study found significant global DNA hypermethylation in adult male rats exposed to both BPA and TFD. METTL14 enhances ZFP14 mRNA stability through m6A modification, leading to increased ZFP14 expression, which in turn promotes STAT3 ubiquitination and degradation. This METTL14/ZFP14 axis suppresses ccRCC growth and metastasis, highlighting a potential therapeutic target for ccRCC treatment.

Li ZL et. al. (February 2025). NSUN2-mediated HCV RNA m5C Methylation Facilitates Viral RNA Stability and Replication Genomics Proteomics Bioinformatics.
This study investigates how NSUN2-mediated m5C methylation in hepatitis C virus (HCV) RNA enhances viral RNA stability and replication. The study shows that NSUN2, induced by HCV infection, facilitates m5C modification at a specific site in the viral genome, promoting viral replication, assembly, and budding. These findings suggest that NSUN2 is a potential therapeutic target for Flavivirus infections.
