Secondary antibodies are crucial reagents used in many research applications, particularly in the field of immunology. They are typically used in conjunction with primary antibodies, which are specific to the target antigen of interest. The primary antibody binds to the antigen, while the secondary antibody binds to the primary antibody and provides a means to detect and quantify the antigen-antibody complex.
Secondary antibodies are used in a wide range of techniques such as Western blot, ELISA, Immunohistochemistry, Immunofluorescence, Flow Cytometry, etc. They are extremely important in these techniques because they allow the detection of low levels of antigens, and they also increase the specificity/sensitivity of the assay.
Secondary antibodies are commonly conjugated to enzymes such as horseradish peroxidase (HRP) or alkaline phosphatase (AP) to facilitate detection, or to fluorescent dyes for use in fluorescence-based techniques. The use of secondary antibodies also allows for multiplexing, where multiple different antigens can be detected in the same sample simultaneously.
