This study identifies a mechanism in early-onset preeclampsia involving the regulation of FAM46C expression by DNA methylation. Downregulation of DNMT3A increases gene body methylation and transcription of FAM46C, which disrupts the migration and invasion of trophoblast cells. Elevated FAM46C expression and gene body methylation were observed in preeclamptic placentas, highlighting a potential epigenetic contributor to the condition.

Huang H et. al. (November 2024). SlVQ15 recruits SlWRKY30IIc to link with jasmonate pathway in regulating tomato defence against root-knot nematodes Plant Biotechnol J.
This study uncovers a mechanism by which tomatoes defend against root-knot nematodes (RKNs). The protein SlVQ15 recruits SlWRKY30IIc, forming a complex that enhances resistance to Meloidogyne incognita without compromising plant growth. The jasmonate (JA) pathway is integral to this defense, with SlVQ15 and SlWRKY30IIc suppressing JA-ZIM domain (JAZ) repressor expression. A feedback loop involving SlMYC2 and SlJAZ5 further modulates this regulatory circuit. These findings highlight potential genetic targets for improving tomato resistance to RKNs.

Deng K et. al. (November 2024).  Ythdf2 facilitates precursor miR-378/miR-378-5p maturation to support myogenic differentiation Cell Mol Life Sci. 81(1):445.
This study reveals that Ythdf2 supports skeletal muscle differentiation by promoting the maturation of pre-miR-378/miR-378-5p in an m6A-dependent manner. Ythdf2 interacts with DICER1 and TARBP2 to enhance the expression of mature miRNAs, which activate the mTOR pathway, facilitating myotube formation. Knockdown of Ythdf2 or miR-378/miR-378-5p inhibits myogenic differentiation, while their overexpression rescues this effect. These findings highlight Ythdf2’s role in regulating myogenesis through miRNA maturation and m6A modification.

Wu W et. al. (November 2024). METTL14 regulates inflammation in ulcerative colitis via the lncRNA DHRS4-AS1/miR-206/A3AR axis Cell Biol Toxicol. 40(1):95.
This study identifies METTL14 as a protective factor in ulcerative colitis (UC) by regulating inflammation through the lncRNA DHRS4-AS1/miR-206/A3AR axis. METTL14-mediated m6A modifications stabilize DHRS4-AS1 transcripts, which reduce inflammatory injury by modulating the miR-206/A3AR pathway. Knockdown of METTL14 exacerbates colonic damage, apoptosis, and inflammatory cytokine production in both cell and murine colitis models. Overexpression of DHRS4-AS1 reverses the inflammatory effects of METTL14 silencing. These findings highlight METTL14 as a potential therapeutic target for UC.

Yu J et. al. (November 2024). ALKBH5 activates CEP55 transcription through m6A demethylation in FOXP2 mRNA and expedites cell cycle entry and EMT in ovarian cancer Biol Direct. 19(1):105.
This study reveals that ALKBH5 promotes ovarian cancer (OC) progression by regulating the FOXP2/CEP55 axis. ALKBH5-mediated m6A demethylation destabilizes FOXP2 mRNA, reducing its ability to repress CEP55 transcription. Elevated CEP55 expression is associated with poor prognosis, enhanced cell proliferation, invasiveness, and epithelial-mesenchymal transition (EMT). Conversely, CEP55 knockdown suppresses tumor growth and metastasis. These findings underscore the role of ALKBH5 in OC progression and suggest targeting the ALKBH5/FOXP2/CEP55 axis as a potential therapeutic strategy.
