In the Colloidal Gold Immunochromatography Assay detection system, the background of antibody (SARS-CoV-2 Spike RBD Recombinant Nanobody) is clean, the detection limit can be as low as 25ng/ml (1. 75ng/0. 07ml), and the sensitivity is very good.

ELISA: Immobilize various types of SARS proteins at a concentration of 2μg/ml on a solid substrate, then react with SARS-CoV-2 Spike RBD Recombinant Nanobody at concentrations of 100μg/ml, 10μg/ml, and 1μg/ml. It shows the nanobody is specific for SARS-CoV-2-S1-RBD protein, without any cross-reactivity with MERS-CoV, SARS-CoV, HCoV-OC43, or HCoV-229E.

Activity Assay – ELISA: Measured by binding ability in a functional ELISA. Immobilized SARS-CoV-2-S1-RBD (2 µg/ml) binds SARS-CoV-2 Spike RBD Recombinant Nanobody with an EC₅₀ of 0.8674 ng/ml.

SARS-CoV-2 Spike protein RBD His/Sumostar Tag captured on a COOH chip binds to the SARS-CoV-2 Spike RBD Recombinant Nanobody, increasing the local refractive index (RI) and causing a red shift of the LSPR peak position. Higher nanobody concentrations produce a larger wavelength shift. The detected affinity constant is 28.2 nM.

SARS-CoV-2 Spike RBD Recombinant Nanobody competed with ACE2-HRP conjugate for binding to SARS-CoV-2-S1-RBD. The binding signal decreased as nanobody concentrations increased, indicating effective inhibition of SARS-CoV-2-S1-RBD/ACE2 binding. The IC₅₀ is 1.296 nM.

SARS-CoV-2 Spike RBD Recombinant Nanobody competitively prevented SARS-CoV-2-S1-RBD from binding to ACE2-HRP conjugate. Inhibition of SARS-CoV-2-S1-RBD/ACE2 binding increased with nanobody concentration, demonstrating effective inhibition. The IC₅₀ is 0.1074 μg/ml.
