BackgroundING1 (Inhibitor of Growth1) is the founding member of a family of five genes (ING1–5). ING proteins are involved in chromatin remodeling, cellular senescence, differentiation, DNA damage response, cell cycle regulation and apoptosis. ING1 interacts with the proliferating cell nuclear antigen (PCNA) in a UV-inducible manner. It also interacts with the 14-3-3 family members leading to its cytoplasmic relocalization. Overexpression of this gene increases expression of the Bax gene. It has also been reported to modify mitochondrial membrane potential in a p53-dependent manner. ING1 is classified as a type II tumor suppressor. While, mutations in this gene are considered unusual, different types of human cancers present diminished levels of ING1 protein and, in some examples, localization in the cytoplasm. Multiple alternatively spliced transcript variants encoding distinct isoforms have been reported for this gene.
